چکیده

Mice genome responses to MTX (anticancer drug) and some plant (Curcuma, Ginger, Green Tee and Pomegranate) extracts were studied. Mice were intraperitoneally injected with 0.75 mg/kg chronic dose of MTX for two weeks. Mortality percentages, bone marrow cell divisions, morphological and biochemical characterization of treated mice groups were estimated. The mortality percentage was 40 % in the positive control while no mortality was observed in both (MTX, Curcumin) and (MTX, Pomegranate) treatments. Some treated mice had ulceration and hair loss on the ears skin. No morphological changes were observed on the negative control group. At cytogenetic level, no bone marrow cell divisions were detected in treated mice. Two Isozyme systems (Esterase and Super oxide dismutase) and Protein electrophoresis were assayed to detect biochemical genetic markers for all mice groups. These analyses reflect mice genome responses under experimental conditions. [Mansour, A.A. M.A. Salam, and Y. M. Saad. Evaluation of effectiveness of some mitochondrial genes in biosystematics and phylogeographic studies of house mouse (Mus musculus ) subspecies

The identification of the efficiency of some mtDNA genes of Mus musculus species complex (house mouse) for biosystematics research was studied in this approach. Recent studies have made use of different mitochondrial genes including NADH dehydrogenase genes, cytochrome b gene, cytochrome oxidase genes, D-loop region and whole mtDNA genome to study the house mouse species. Usage of each of these...

Karyotype of NIH, C57BL/6 and Razi strains of laboratory mice (Mus musculus)

The laboratory mouse is recognized as the pre-eminent model for genetic research. Awareness of chromosomal patterns of experimental animals increases their value for a variety of different fields of study. We aimed to study mitotic chromosome preparations from NIH, C57BL/6 and Razi strains of mice, which are outbred, inbred and partially inbred laboratory mice respectively. Bone marrow cells we...

Pharmacokinetics and Biodistribution of Pegylated Methotrexate after IV Administration to Mice

The efficacy of methotrexate (MTX) as an antimetabolite chemotherapeutic agent highly depends on its blood circulation half-life. In our previous study, different conjugates of MTX (MTX-PEG) were synthesized, their physicochemical properties were investigated and MTX-PEG5000 was finally selected as optimum drug-conjugate for further investigations. In the current work, first the stability of MT...

Protective effects of cornus mas extract on in vitro fertilization potential in methotrexate treated male mice

Current study was aimed to evaluating protective effects of cornus mas fruit extract (CMFE) in mice treated with methotrexate (MTX). For this purpose, 48 young mature male mice were divided into 6 groups. Control group received only normal saline (0.1 mLper day, intraperitoneally), and the second group was administered MTX (20 mg kg-1 perweek, intraperitoneally). The third, ...

Protective effects of vitamin E and Cornus mas fruit extract on methotrexate-induced cytotoxicity in sperms of adult mice

This study was aimed to assess the protective effects of Cornus mas fruit extract (CMFE) and vitamin E (Vit E) on sperm quality parameters in the methotrexate (MTX)-treated mice. Forty-eight young adult male mice (8-12 weeks) were randomly divided into six groups including control and test groups. The control group received normal saline orally , and the test groups were treated MTX (2...
